The answer to this question depends on a lot of things such as the architecture of the application, platform support, tester skills and knowledge, budget for tolls, etc. Also, which tool is best is rather subjective.

-Web Developer Extension for Firefox
-Selenium- A suite of open source web automation tools
-Fiddler- An HTTP/HTTPS proxy
-Watcher- A Fiddler extension
-PICT- A combinatorial test tool from Microsoft
-Fireeye/ACTS- Another combinatorial test tool from NIST
-Jedit- Open source text editor with a great set of available extension.
