The ipspoof.dat file on a particular player needs to contain all ips you want to spoof from that player. In order to spoof an ip, it must be statically bound to the NIC card on that player. This means that when you look at the ipconfig from the command line every ip address you intend to use should show up.
If you check the "Generate IP Spoof Data" option under the player machine configuration in Conductor the ipspoof.dat file will automatically be created when your test is launched and will contain all statically bound ips on that machine. Note that even with this option checked if an ipsoof.dat datapool exists in the datapools folder on the player machine it will not be overwritten.
