Generating GUI checklist primarily before application testing for particular version
Is it necessary to generate a GUI checklist before generating the WinRunner script for a particular version of the application ?
Is there any use of this checklist for the later versions of the applications ?

1st question answer: No
2nd question answer: Yes. If the applications under test haven't changed in such a way to negate your checklist, they should still be applicable. The checklists are an important way to detect regression defects and are good at letting you know when the apps change within the scope of the checklists.
My yet unanswered question to you:
Why are you using 6.0x?
