This scenario should not be an issue.
As I understand it - when you run a manual test QC copies the steps from Test Plan to the test Lab test. So no locks are held on the Test Plan test.
Similarly with Automated test scripts like QTP/WinRunner - if you are running tests from different test sets in the test lab that are all instances of the same test in test plan then you execute copies of the test plan test and save the results of the tests to the test in the test lab.
In this instance you will find that you can only open the test in read only mode on the second machine. But all this means is that you can not save any changes to script. You can still run it fine.
