i'm going to guess that someone asked you do do this or look into this -- you might want to talk to them first in order to find out what they meant (or what tools they might already have).
my guess would be to use some sort of tool to monitor the "usage" on the server CPU (or CPUs) while you exercise a variety of tests against that server. it may also involve logging CPU usage on each client, too.
This might be something as simple as logging the output from a MMC\Performance (Windows) or a "top" (unix) session. A tool like sitescope might be useful in sniffing out which transaction is causing the response on the server.
when trying to target performance issues, it's important to be simulating as realistic a production environment as possible. things like connection speed, test v. production server capacities, etc. will result in different response profiles.
